Minimum Essential Medium in Earle’s Buffer-Premium (MEM–EBSS-Premium)

MEM–EBSS-Premium is formulated with L-Alanyl-L-Glutamine, providing an extended shelf life. MEM–Premium in EBBS (Earle’s Balanced Salt Solution) substantially enhances cell viability and proliferation. GMP Bioscience™ offers multiple MEM–EBSS-Premium variants tailored to your specific needs.

Background

MEM–Premium in Earle’s Buffer is a widely utilized medium for culturing mammalian cells.

Advantages of MEM–EBSS-Premium Compared to Standard MEM-EBSS
– Increased pH buffering capacity
– Contains 4 mM L-Alanyl-L-Glutamine (0.869 g/L) replacing conventional L-glutamine,

which:

– Extends shelf stability
–  Reduces the accumulation of toxic ammonia
– Improves cell viability and growth rates
– Maintains stability over a broad temperature range
–  Enhances the culture of primary cells

Composition

MEM–Premium in Earle’s Buffer includes:

– Sodium pyruvate: 5 mM
– High glucose: 4.5 g/L
– Non-essential amino acids
– Sodium bicarbonate: 3.7 g/L
GMP Bioscience™ manufactures several MEM–EBSS-Premium formulations varying in sodium bicarbonate and HEPES concentrations for optimized pH control

– Phenol red as a pH indicator

MEM–Premium in EBBS contains no proteins, lipids, or growth factors and therefore requires supplementation, typically with 10% fetal bovine serum (FBS) for most cell culture applications.

Mechanism:

While L-glutamine is an essential amino acid, it tends to break down over time, leading to the formation of harmful byproducts such as ammonia and pyrrolidine carboxylic acid. One strategy to reduce its degradation in culture media is by slowly supplementing it throughout the culture period.

However, consistently monitoring and maintaining optimal levels can be labor-intensive and impractical.

A more efficient solution is to use L-alanyl-L-glutamine, which offers significantly greater stability in aqueous environments. Unlike free L-glutamine, it resists degradation and releases aminopeptidases at a controlled rate. This results in a sustained supply of L-glutamine and L- alanine, which are then utilized by cells for protein synthesis and energy metabolism via the TCA cycle.
