Roswell Park Memorial Institute 1640-Premium (RPMI-1640-Premium) contains L-Alanyl-L- Glutamine, which supplies a longer shelf life.  RPMI-1640-Premium significantly improves cell viability and growth, and is mainly used for mammalian cell culture.

Mechanism

L-glutamine is an essential amino acid, but it is prone to breakdown, leading to the formation of harmful byproducts such as ammonia and pyrrolidine carboxylic acid. One approach to limit its degradation in culture media is by slowly supplementing the medium with fresh L-glutamine over time. However, maintaining precise timing and concentration levels is laborious and often impractical.

As an alternative, L-alanyl-L-glutamine offers a more stable option in aqueous environments. It resists rapid breakdown and releases aminopeptidases in a controlled manner. This provides a steady supply of L-glutamine and L-alanine, which cells utilize for protein synthesis and energy metabolism via the TCA cycle.
